- Web search for the human gene, e.g. CACNA1A – information on function, location, etc.
- Web search for human protein sequence: click on UniProt link or go directly to uniprot.org
- Scroll down to “Sequences” and choose “FASTA” (Sequence data in FASTA format). Copy the text sequence.
- Go to wormbase BLAST and paste the human protein sequence. Select Query Type: protein and OK.
- Locate genes with highest alignment, greatest Score (bits), and an E-value of 0.0
- In this example, the gene unc-2 has:
- Score 901 bits, E-value = 0, Identities = 439/767 (57%), Positives = 558/767 (72%)
- Click [Gene Summary] to go to the WormBase gene, unc-2 to verify that the reported worm gene function matches the human gene function
- Click [Alignment] to view exact amino acid matches and mismatches
